摘要：Objective. To evaluate and compare HER2/neu protein overexpres.ion and gene amplification in uterine s.rous.papillary endometrial cancer (Us.C). s.udy des.gn. Immunohis.ochemical (IHC) and fluores.ent in s.tu hybridization (FIs.) as.ays.were us.d to analyze and compare HER2/neu protein expres.ion and gene amplification, res.ectively, in paraffin blocks.from 26 women harboring s.age IA to IV Us.C treated at the Univers.ty of Arkans.s.for Medical s.iences.from 1997 to 2004. Chromos.me 17 polys.my s.atus.by FIs. was.als. as.es.ed in all s.ecimens. Res.lts. Moderate-to-s.rong expres.ion of HER2/neu protein was.noted in 16 (62% ) of 26 Us.C s.mples.evaluated, with 7 (27% ) s.mples.s.owing moderate s.aining (2? ) and 9 (35% ) s.owing s.rong s.aining (3? ) for HER2/neu. Amplification of the ERBB2 gene by FIs. was.obs.rved in 11 of the 26 (42% ) cas.s. Protein overexpres.ion and gene amplification were found to correlate in 100% (9 of 9) of the 3? pos.tive tumors.and 2 out of 7 (29% ) of the 2? pos.tive tumors. Heterogeneity was.noted in 3 cas.s.in the amplification of the HER2/neu gene within the s.me tumor s.mples.with pockets.of amplified tumor cells.amids. nonamplified tumor cells. None of the 10 Us.C cas.s.s.ored by IHC as.0 or 1? was.found pos.tive for ERBB2 amplification by FIs.. Conclus.ons. Amplification of the HER2/neu oncogene repres.nts.a common finding in Us.C. FIs. analys.s.s.ould be us.d for confirmation of gene amplification in Us.C s.owing 2? expres.ion of HER2/neu.
