摘要：AIM To investigate the enhanced cytotoxic T lymphocyte responses against pancreatic cancer (PC) in vitro induced by dendritic cells (DCs) engineered to secrete anti-DcR3 monoclonal antibody (mAb). METHODS DCs, T lymphocytes and primary PC cells were obtained from PC patients. DCs were transfected with a designed humanized anti-DcR3 monoclonal antibody heavy and light chain mRNA and/or total tumor RNA (DC-tumor-anti-DcR3 RNA or DC-total tumor RNA) by using electroporation technology. The identification, concentration and function of anti-DcR3 mAb secreted by DC-tumor-anti-DcR3 RNA were determined by western blotting and enzyme-linked immunosorbent assay. After co-culturing of autologous isolated PC cells with target DCs, the effects of secreting anti-DcR3 mAb on RNA-DCs' viability and apoptosis were assessed by MTT assay and flow cytometry. Analysis of enhanced antigen-specific immune response against PC induced by anti-DcR3 mAb secreting DCs was performed using a Cr-51 releasing test. T cell responses induced by RNAloaded DCs were analyzed by measuring cytokine levels, including IFN-gamma, IL-10, IL4, TNF-alpha and IL-12. RESULTS The anti-DcR3 mAb secreted by DCs reacted with recombinant human DcR3 protein and generated a band with 35 kDa molecular weight. The secreting mAb was transient, peaking at 24 h and becoming undetectable after 72 h. After co-incubation with DCtumor- anti-DcR3 RNA for designated times, the DcR3 level in the supernatant of autologous PC cells was significantly down-regulated (P < 0.05). DCs secreting anti-DcR3 mAb could improve cell viability and slow down the apoptosis of RNA-loaded DCs, compared with DC-total tumor RNA (P < 0.01). The anti-DcR3 mAb secreted by DC-tumor-anti-DcR3 RNA could enhance the induction of cytotoxic T lymphocytes (CTLs) activity toward RNA-transfected DCs, primary tumor cells, and PC cell lines, compared with CTLs stimulated by DC-total tumor RNA or control group (P < 0.05). 摘要：材料技术发展一直备受科技界和产业界的关注。采用一种多相材料水平集拓扑形状优化方法,对具有周期性微结构的人工复合材料进行微结构拓扑优化设计,使复合材料具有极端的热弹性能。这一系统的数值方法,通过均匀化方法计算微结构的等效材料性能,然后采用基于多相水平集模型的参数化水平集方法实现微结构的形状和拓扑演变。多相水平集模型使用多个水平集函数的组合来描述多相材料的边界,并且不出现重叠和缺失。参数化水平集方法将汉密尔顿-雅各比偏微分方程的拓扑形状优化问题转换为一个较为简单的尺寸优化问题。由两个极限热弹性材料微结构设计算例,验证了该设计方法的可行性,为设计具备极限性能的人工材料提供了一套系统的设计方法。

摘要：针对传统的机器阅读理解模型难以胜任从多篇文档中学习多跳实体间语义关系的多跳阅读理解任务,提出一种基于多视角的图卷积神经网络模型MV-GCN。受Inception启发,通过构建多通路的图卷积来拓宽网络,提高模型多视角学习多跳实体间语义相关性的表达能力。引入基于Squeeze-and-Excitation的多视角注意力机制实现前述多视角表达的有效融合。在Wiki Hop通用数据集上的实验结果表明,MV-GCN在多跳阅读理解任务上取得了很好的效果,其性能优于当前的主流方法,可为进一步研究提供借鉴。

摘要：Li-metal anodes are one of the most promising energy storage systems that can considerably exceed the current technology to meet the ever-increasing demand of power applications. The apparent cycling performances and dendrite challenges of Li-metal anodes are highly influenced by the interface layer on the Li-metal anode because the intrinsic high reactivity of metallic Li results in an inevitable solid-state interface layer between the Li-metal and electrolytes. In this review, we summarize the recent progress on the interfacial chemistry regarding the interactions between electrolytes and ion migration through dynamic interfaces. The critical factors that affect the interface formation for constructing a stable interface with a low resistance are reviewed. Moreover, we review emerging strategies for rationally designing multiple-structured solid-state electrolytes and their interfaces, including the interfacial properties within hybrid electrolytes and the solid electrolyte/electrode interface. Finally, we present scientific issues and perspectives associated with Li-metal anode interfaces toward a practical Li-metal battery.

摘要：某锅炉用楔形双闸板闸阀在实际使用过程中出现了闸板密封面磨损严重、阀体上导轨槽与闸板上导轨翻边现象,这会对阀门密封面及其整体结构造成很严重的破坏,影响使用寿命。结合尺寸链理论对导轨间隙尺寸进行分析,对导轨槽挤压应力进行校核,并基于热固耦合仿真分析方法对其进行模拟。结果表明:由于导轨间隙尺寸过大,闸板提起时不能及早使两密封体完全脱离产生磨损,导轨结构尺寸的不合理使其导轨上挤压应力不满足要求,在长期高压高温工况使用下,阀体导轨槽产生屈服被压溃,致使闸板倾斜挤压密封面,造成进一步的磨损破坏。根据研究结果对闸板上导轨结构尺寸进行优化改进,满足了所需应力要求,减小磨损程度。此研究可为该类阀门的设计及优化提供一定的参考依据。

摘要：装配信息建模是数字化装配、智能化装配的基础,然而人工交互建模效率低下且易发生装配特征识别和装配特征配合的错误,难以满足复杂机械系统的精确建模。基于三维装配体模型隐含的装配工艺信息,以STEP模型文件为输入,研究了面向几何信息的装配特征自动提取与配合关系识别算法。进而针对装配集成信息模型的构建,从装配精度模型和装配序列规划角度提出了信息推理算法。最后基于开发的系统,通过装配实例信息模型构建证明了所提算法的有效性。
