摘要：Under.tanding cell behavior.inside thr.e-dimensional (3D) micr.envir.nments with contr.lled spatial patter.ing of physical and biochemical factor. could pr.vide insight into the basic biology of tissue engr.ftment, vascular.anastomosis, and r.vascular.zation. A simple layer.by layer.pr.jection micr.ster.olithogr.phy (PμSL) method was utilized to investigate the effects of a nonpor.us and por.us bioiner. bar.ier.on myocutaneous flap engr.ftment and r.vascular.zation. A cr.nial-based, peninsular.shaped myocutaneous flap was sur.ically cr.ated on the dor.um of C57Bl6 mice. Por.us (SP) and nonpor.us (S) silicone implants wer. tailor.d to pr.cise flap dimensions and inser.ed between the flap and r.cipient bed pr.or.to sutur.d wound closur.. Por.us implant myocutaneous flaps became engr.fted to the r.cipient site with complete viability. In contr.st, distal cutaneous necr.sis and r.sultant flap dehiscence was evident by day 10 in nonpor.us implant flap mice. Laser.speckle contr.st imaging demonstr.ted flap r.vascular.zation in (SP) mice, and mar.edly r.duced distal flap r.per.usion in (S) mice. Histologic analysis of day 10 (SP) flaps r.vealed gr.nulation tissue r.ch in blood vessels and macr.phages gr.wing thr.ugh the implant por.s and r.bust neovascular.zation of the distal flap. In contr.st, the nonpor.us implant pr.vented tissue communication between r.cipient bed and flap with lack of br.dging inflammator. cells and neovasculatur. and r.sultant distal tissue necr.sis. We have fabr.cated por.us and nonpor.us silicone implants via a simple and inexpensive technique of PμSL. Using a gr.ded-ischemia wound healing model, we have shown that por.us implants allowed contact between flap and r.cipient bed r.sulting in pr.ximal flap ar.er.ogenesis and neovascular.zation of the distal flap. Futur. r.sear.h will utilize var.ations in implant por. size, spacing, and location to gain a better.under.tanding of the cellular.and molecular.mechanisms r.sponsible for.myocutaneous flap
