摘要：The ecological costs of open pit metal mining are quantified, which include lost value of direct eco-services, lost value of indirect eco-services, prevention and restoration costs, and cost of carbon emission from energy consumption. These ecological costs are incorporated in an iterative ultimate pit optimization algorithm. A case study is presented to demonstrate the influence of ecological costs on pit design outcome. The results show that it is possible to internalize ecological costs in mine designs. The pit optimization outcome shifts considerably to the conservative side and the profitability decreases substantially when ecological costs are accounted for.

摘要：抗生素的滥用使得大量抗生素进入污水处理厂中,但乡镇污水处理厂中抗生素的污染尚未引起重视.为了解乡镇污水处理厂中抗生素的污染水平及去除特性,考察了15座乡镇污水处理厂进出水中15种抗生素的污染水平和去除效率,并评估了其对水生生物和人体健康构成的风险.结果表明,进出水中四环素类抗生素(TCs)检出浓度最高,四环素(TC)和土霉素(OTC)最高检出浓度可达(4 943.69±4.31)ng·L^(-1)和(3 907.81±52.04)ng·L^(-1).在乡镇污水处理厂中,A/O和A^(2)/O工艺对抗生素的去除能力强于MBR工艺.生态风险评估表明,TCs和喹诺酮类抗生素(FQs)具有较大的风险商值,会对水生生物造成中高风险;而磺胺类抗生素(SAs)对水生生物造成低风险.虽然抗生素会对水生生物造成风险,但对人体健康的风险可忽略不计.研究结果可为今后乡镇污水处理厂工艺的升级改造和地区抗生素排放标准的制定提供支撑.

摘要：【目的】环氧化物水解酶(epoxide hydrolases,EHs)在手性药物的合成中起着重要作用。为了补充和发现更多的高性能环氧化物水解酶,通过基因调取技术探索新的环氧化物水解酶。【方法】通过基因调取技术鉴定了一种来自卡尔斯巴德曲霉的新型环氧化物水解酶(Aspergillus carlsbadensis epoxide hydrolase,Ac EH)。采用Auto Dock2预测Ac EH的关键水解位点,通过计算设计阐明重要位点对Ac EH的结构和催化机制的影响。【结果】对新型Ac EH一级结构的分析揭示了3个特征性α/βEH基序的存在:HGWP、GYTFS和GGDIGS。Ac EH酶表现出高活性,可在15 min内完全水解氧化苯乙烯(styrene oxide,SO),比活性为13951 U/g。Km、V_(max)和k_(cat)/K_(m)值分别为(107.07±57.98)mmol/L、(37.22±17.85)μmol/(min·mg)和1.17 mmol/(L·s)。Ac EH的关键水解位点是催化三联体的Asp192-His372-Glu346和2个保守酪氨酸Tyr251/314。某些突变(R49L、R49Y)导致酶失活,而其他突变(Y45L)导致无活性包涵体的形成。相互作用网络分析显示,第49个氨基酸残基的变化破坏了重要活性位点残基的相互作用,导致酶失活。另一方面,第45个氨基酸残基的改变使酶的结构不稳定,导致包涵体的形成。【结论】本研究发现了一种新的环氧化物水解酶,并分析了其水解机制,为进一步研究这种酶及其工业化应用提供了有价值的见解。

摘要：Based on the lead compound 1 reported in literature, a series of novel BACE1 inhibitors were designed and synthesized, among which compound 11 exhibited a 14-fold improvement in potency over the lead compound 1. This represents a good lead for the discovery of more promising BACE1 inhibitors for the potential treatment of AD.

摘要：By taking compound 1 as a lead, a series of 5-cyclopropyl substituted cyclic acylguanidine compounds were designed and synthesized as BACE1 inhibitors, compound 4d exhibited 84-fold improved inhibition efficiency than lead compound 1. The diphenyl fragment at the P3 position and the substituents at the second phenyl ring were essential for the compounds to achieve improved inhibition efficiency. This SAR studies provides new insights into the design and synthesis of more promising BACE1 inhibitors for the potential treatment of AD.

摘要：本文以正在建设的1.5级跨音压气机试验台所采用的压气机为研究对象,利用NUMECA软件计算了该压气机在不同转速下的特性曲线,并在设计转速下,对其内部流场进行了分析研究。研究表明:激波位置和强度随压气机运行工况的变化而变化;在近失速工况和堵塞工况时,压气机内部流动出现分离;在设计工况时,压气机内部流动状况良好。上述结论,可以为试验台建设中,探头位置的确定和激光测窗位置的选取等提供指导和帮助。此外,该算例作为一盲解,将来同实验结果对比后,可以用来检验和校准CFD程序,为国内F级燃机设计平台的建设打下良好的基础。

摘要：利用粒子经典理论建立了自放大自发辐射(SASE)自由电子激光高增益自洽方程组,编写了稳态Matlab-SASE-FEL数值计算程序;对LCLS装置参数进行了优化,给出了辐射光的各特征参数。该程序的主要优势在于基于Matlab的便利性可方便地进行后处理数据,对X射线FEL理论研究的深化和实验装置的设计和改进具有重要的意义。
