摘要：[Objective] This study aimed to establish a multiplex PCR system for de- tecting transgenic ingredients from Citrus. [Method] Based on the pBI121 plasmid sequences published in GenBank and actin gene sequence of Citrus, the primers specific to CaMV35S promoter, NOS promoter, NOS terminator and actin gene were designed, to establish a multiple PCR system which could detect four types of sequences. In addition, orthogonal tests were performed to determine the optimal concentrations of all the components in PCR reaction system, as well as the optimal PCR cycle parameters. [Result] The optimal PCR reaction system should contain 2.5μl of 10xPCR buffer, 2.0μl of MgCI2 （25 mmol/L）, 2.0 μl of dNTP mixture （2.5 mmol/L of each dNTP）, 1.0 μl of actin gene primers （10μmol/L）, 1.0μl of 35S promoter primers （10 μmol/L）, 1.5 μl of NOS promoter primers （10 μmol/L） and 0.5 μl of NOS terminator primers （10μmol/L）, 0.1 μg of template DNA, 1.25 U of Taq DNA polymerase; ddH20 was added to the total reaction system of 25μl. The PCR reaction program consisted of pre-denaturing at 94℃ for 5 min; 31 cycles of denaturing at 94℃ for 30 s, annealing at 64.1℃ for 45 s and extension at 72℃ for 50 s; final extension at 72℃ for 10 min. The reaction system optimized with the orthogonal tests could detect as less as 0.1% transgenic component in the tested samples. [Conclusion] The MPCR detection system established in this study can meet the requirements in theory for detecting the genetically modified ingredients in Citrus or the deep-processed products.

摘要：[Objective] The large-scale single-column fattening pig house with fermen- tation bed could hold 1 500 heads of fattening pigs. Since the number of pigs in piggery is too large, the management is difficult. The behavior of feeding, drinking, movement, sleeping, fighting of pigs is difficult to handle. The pigs cannot be man- aged well, resulting in the enhanced weakness of piglets, enhanced illness of weak pigs and missing treatment of ill pigs. The management for the pig populations is not satisfactory, and thus, it is needed to improve timely. [Method] The barriers for the fattening pigs in the large-scale pig house with fermentation bed were designed. The single management for single fattening pig was proposed. The large-scale fat- tening pig house was divided into 8 regions. Among them, 4 regions were located in both sides of the fermentation bed. Their main function was to separate ill, weak, small and bad pigs. In addition, the main column was divided into 4 gradual barri- ers. They were used to separate different-size fattening pigs. In view of manage- ment, the different-type pigs were managed dividedly with the gradual barriers. The equally-sized pigs were concentrated into one column. The ill, weak, small and bad pigs were isolated into barriers. Thus, the dynamic management was adopted. Until the fattening pigs grew up to 75 kg and their health was stable, the barriers among the columns were canceled to mix the pigs again and guarantee the pigs more gymnastic space. [Result] This design would improve the disease resistance of ill pigs, health status of weak pigs and management level of pig populations. This study would also provide a basis for the healthy running of large-scale fattening pig house with fermentation bed. [Conclusion] The pig-raising model with fermentation bed would improve the environment of pig house and the welfare of pigs. 摘要：[Objective] The aim was to provide a theoretical basis for stable and highly effective intercropping arrangements and scientific management measures by selecting apple, pear, peach, apricot, walnut, jujube and other fruit trees to study their influence on yield, fiber quality and economic returns of intercropped cotton in southern Xinjiang. [Method] Based on major cropping pattern in production, randomized block design was adopted to explore growth indicators, canopy micrometeorological indicators, yield and fiber quality in key growth stage. [Result] Shading has a significant effect on cotton canopy micro-environment and canopy diameter is proportional to shading effect. According to comparisons of the same tree type, the change of canopy micro-environment was as follows： under canopyouter canopymiddle points and peachpearapplewalnutjujube for comparisons among different tree types. Canopy diameter is directly proportional to the number of tree branch and boll weight reductions and shading is the main cause of yield reduction. The canopy expansion is the major cause of decline of light intensity, temperature and humidity of cotton canopy. [Conclusion] Fruit trees, which will promote cotton yield,quality and canopy-environment, are as follows： jujube walnut apple pear peach trees. In practice, trees, which are small in canopy or well trimmed, are popular in production, such as jujube trees, to improve cotton yield and fiber quality.

摘要：[Objective] In this study, the quantitive detection of PCV2 （porcine circovirus type 2） in vitro was achieved. We aimed to establish two kinds of TaqMan real-time PCR methods based on PCV20RF1 and ORF2 respectively and compare them. [Method] According to the relatively＇conserved sequences of PCV20RF1 and ORF2 registered in GenBank, two pairs of specific primers and TaqMan probes were designed and synthesized. Then the recombinant plasmids containing the whole sequences of PCV20RF1 and ORF2 were constructed to draw the standard curves through optimizing the reaction system and conditions. And thus two kinds of TaqMan real-time PCR detection methods based on the whole sequences of ORF1 and ORF2 respectively were constructed for PCV2. [Result] For the two established standard curves, the Ct values showed a good linear relationship with the loga- rithms of copy numbers of templates （F2〉0.99）. The amplification efficiency ranged from 90% to 110%. The amplifications all had a good repeatability with variation coefficients within groups all less than 5%. Moreover, the amplifications all had a good specificity. When the sequences of porcine parvovirus （PPV）, porcine circovirus type 1 （PCV1）, swine pseudorabies virus （PRV）, porcine reproductive and respiratory syndrome virus （PRRSV） were used as templates, the target sequence was not amplified. The amplifications also had a high sensitivity. The ORF1 detection method could reach 1.0x10T copies/;ul, and the ORF2 detection method could reach 1.0×10^2 copies/μl. The two established real-time PCR detection methods were used to detect the 80 clinical samples respectively. The results showed the magnitudes of 72 amplified samples were basically consistent between the 2 detection methods, while the magnitudes of the other 8 amplified samples were inconsistent. 摘要：Objective] This study aimed to determine the effects of medium composi-tion and fermentation condition on the production of antibiotics from an actinomycete antagonistic to Penicil ium viridicatum, which was isolated from soil and identified primarily as Streptomyces anandi . [Method] Single-factor experiments and orthogonal array designs were used to optimize the medium composition and fermentation con-ditions. [Result] Culture medium composition and fermentation conditions showed sig-nificant influence on the antibiotic production from S. anandi . The optimum culture medium consisted of soluble starch 10 g/L, soybean cake powder 15 g/L, KH2PO4 0.5 g/L and MgSO4·7H2O 0.1 g/L. The optimized fermentation conditions were tem-perature 30 ℃, initial pH value 7.7, culture medium amount 40 ml/250 ml flask, and fermentation time 144 h. The titer of fermentation broth reached 452.7 U/ml when the S. anandi was cultured under the optimal fermentation medium and conditions. [Conclusion] The study may lay the foundation on further isolation and purification of the antibiotic from S. anandi JXNU02.

摘要：[Objective] This study was conducted to analyze the difference in the general combining ability of yield traits of rice varieties in China and South Asia and to provide reference for the introduction and utilization of rice restorer lines from South Asia. [Method] A total of 111 hybrids were obtained by crossing 37 restorer lines in China and South Asia with Gang 46A, Lu 98A and Dexiang 074 following a NC II design. The combining ability of eight yield traits of the hybrids was analyzed in Luzhou and Deyang bases of Sichuan Province. [Result] In both bases, the eight yield traits had significant or extremely significant differences between the combina- tions, restorer lines, sterile lines or sterile line^restorer line, suggesting that there were d^erences in the genetic effects between the combinations or lines. The gen- eral combining ability （GCA） variances were all higher than special combining ability （SCA） variances in parents, which indicated that these traits were affected by gene additive effect. Restorer lines contributed more to F1 generation than male sterile liens in the eight yield traits. Nine restorer lines from India and Bangladesh were ranked in the top 15 lines with high GCA in the number of productive tillers in both bases. [***] There was significant differences in the combining ability in the eight yield traits between the restorer lines from China and South Asia. The lines from India and Bangladesh were better than those from China in the GCA of the number of productive tillers, and the lines from China were better than those from India and Bangladesh in the GCA of other traits.
