摘要：Field investigation shows that neighboring green space is an important part of the public landscaping for all residential districts,because it is able to highlight effects of landscaping and beautification,satisfy needs of human-centered designs and show individualized landscape characteristics,moreover,it shows great influence on the landscape quality of residential *** view of problems in landscape design of residential districts,this study focuses on plant furnishing patterns,landscape demonstration effects and landscape differences,and then gives suggestions correspondingly,for example,plant furnishing patterns and cultural connotations of landscapes should be improved,human needs put on the top priority,and spaces organized in a proper *** study aims to provide useful reference for the construction of external environment in residential districts.

摘要：Environmental and ecological problems in the urban-rural integration were analyzed, characteristics of urban green space system introduced, such as excellent landscape eco-structure, distinctive historical and humanistic features, and diversified natural landscape resources, and also challenges pointed out. Evolution, overall spatial structure and conservation concept of conservation-minded urban green space system planning were elaborated, on the basis of which the evaluation system of conservation-minded urban green space system was studied, and it was proposed that mutual relationship between impact factors of compound value and qualitative evaluation factors should be taken into consideration as a whole in evaluating conservation effects of green space system. Quantitative evaluation indexes and qualitative evaluation measures were analyzed, an objective and precise evaluation system for the conservation-minded urban green space system was established by combining qualitative and quantitative analysis to improve ecological environment during the urbanization, and fully show planning concepts of conservation-minded green space system.

摘要：The accelerating urbanization has brought various problems in environment, land, fund and so on. With the guidance of market economy, economic urban green spaces have become the optimal choice in urban landscaping of China, which can not only improve urban landscapes, ecological environment, satisfy higher spiritual and cultural demands of people, but also create considerable economic benefits, and enhance the coordinated development of economy, society and environment. With Shijiazhuang City as an example, the approaches of constructing economic urban green spaces fit for Shijiazhuang City were analyzed in this study, to provide references for the construction of economic urban green spaces in other cities.

摘要：根据番茄褪绿病毒(Tomato chlorosis virus,ToCV)热激蛋白70(Hsp70)的基因序列,设计ToCV实时荧光定量PCR特异引物。利用重组质粒ToCV-1为标准品建立SYBR green Ⅰ实时荧光定量方法。针对引物浓度、退火温度、特异性、灵敏度、重复性和稳定性进行系列优化。结果表明,最适退火温度为63℃,最适引物浓度为0.3μmol·L^(-1)。熔解曲线为特异性单峰,表明其特异性良好。建立的SYBR green Ⅰ实时荧光定量PCR较常规PCR灵敏100倍,且具有良好的重复性和稳定性。基于SYBR green Ⅰ实时荧光定量PCR技术建立的ToCV检测方法,速度快、特异性强、灵敏度高、重复性好,可以用于ToCV的定量检测。

摘要：参照GenBank登录的相关基因序列,设计了2对引物分别用于扩增伪狂犬病毒(PRV)gH基因与猪圆环病毒2型(PCV2)ORF2基因的部分片段。将测序正确的PRV gH基因与PCV2ORF2基因片段克隆入pGEM-T Easy载体,转化大肠杆菌DH5α,经测序鉴定后得到阳性重组质粒,作为标准品模板建立SYBR greenⅠ荧光定量PCR标准曲线和熔解曲线,并对其灵敏性、特异性和重复性进行验证。结果显示,PCV2与PRV荧光定量PCR的标准曲线的Tm值分别为80.8℃和86.7℃,熔解曲线特异,灵敏度分别可达215拷贝/μL和180拷贝/μL,是普通PCR检测方法的100倍。结果表明,建立的PCV2与PRV荧光定量PCR检测方法实现了2种病毒的同时检测,能够对PRV、PCV2混合感染的临床病料进行快速诊断。

摘要：Town-and-city concentration area is confronted with ecological degeneration due to the expansion of urbanization,and enriching urban bio-diversity is conducive to the improvement of ecological function of urban green *** paper has studied how to increase urban bio-diversity by making use of urban green lands for protection and construction of wild animals' habitats,and proposed the significance and theoretical idea for protection and construction of wild animals' *** taking ecological green lands design in Qinglongchang of Chengdu City for example,it has analyzed the current situation of animals' diversity;proposed to plan corridor,construct biological channel and provide multiporous habitats;listed plants' selection and collocation mode;and finally illustrated how to set artificial facilities for attraction of *** hopes to provide a reference for the design of ecological green lands in cities advocating the harmonious development of living environment and natural environment.

摘要：根据GenBank中登录的隐孢子虫小亚基rRNA基因序列设计1对引物,并以标准基因组DNA为模板,初步建立了检测乳牛微小隐孢子虫和安氏隐孢子虫的SYBR green real time PCR方法,并对乳牛微小隐孢子虫和安氏隐孢子虫阳性样品和上海40份乳牛粪便进行了检测。结果表明,此次建立的real timePCR对微小隐孢子虫和安氏隐孢子虫均能扩增出曲线,且其他寄生虫(鸡贝氏隐孢子虫、刚地弓形虫、犬新孢子虫)和大肠杆菌均未检测到;标准基因组DNA的检测阈值达到5个拷贝,牛粪中卵囊的最低检测量为每克粪便5个卵囊,乳牛粪便阳性率为15%(6/40)。表明,建立的荧光定量PCR快速、特异、敏感,可用于乳牛隐孢子虫病的流行病学调查。

摘要：旨在建立一种检测马Toll样受体(TLRs)mRNA表达水平的SYBR greenⅠ荧光定量RT-PCR(Rverse Transcription-Polymerase Chain Reaction)方法。参照GenBank中马TLRs的基因序列保守区设计特异性引物,以马β肌动蛋白(β-actin)mRNA为内参,建立荧光定量RT-PCR方法。结果,扩增曲线在1×102～1×108copies.μL-1范围内有很好的线性关系,扩增相关系数在0.990以上,扩增效率在1.0左右。熔解曲线分析表明,产物为特异单峰,无引物二聚体,具有较高的特异性和灵敏度。重复性结果表明,该方法的组内、组间变异系数均小于3.50%,重复性较好。临床样品检测结果表明,TLR1、TLR2、TLR3、TLR4、TLR6、TLR7、TLR8和TLR9在骨髓中均有表达,其中TLR4mRNA表达水平最高,TLR9mRNA表达水平最低。本研究建立的检测方法能够成功用于临床样品的检测,为研究马匹TLRs在mRNA水平的定量分析提供技术平台。
