摘要：The dual-retrieval （DR） operation sequencing problem in the flow-rack automated storage and retrieval system （AS/RS） is modeled as an assignment problem since it is equivalent to pairing outgoing unit-loads for each DR operation. A recursion symmetry Hungarian method （RSHM）, modified from the Hungarian method, is proposed for generating a DR operation sequence with minimal total travel time, in which symmetry marking is introduced to ensure a feasible solution and recursion is adopted to break the endless loop caused by the symmetry marking. Simulation experiments are conducted to evaluate the cost effectiveness and the performance of the proposed method. Experimental results illustrate that compared to the single-shuttle machine, the dual-shuttle machine can reduce more than 40% of the total travel time of retrieval operations, and the RSHM saves about 5% to 10% of the total travel time of retrieval operations compared to the greedy-based heuristic.

摘要：[ Objective] To clone the porcine interleukin-18（/L-18） cDNA and explore the immunological effectiveness of porcine IL-18 as an adjuvant of genetic vaccine. [ Method] The spleen lymphccytes were isolated from Henan three-way cross-breeding pigs. According to the porcine IL-18 gene in GenBank, a pair of specific primers was designed. The full length cDNA of porcine IL-18 was amplified by RT-PCR. Subsequently, porcine IL-18 cDNA was cloned into pGEM-T vector and sequenced and analyzed. [ Result] The porcine IL-18 gene demonstrated an open reading frame of 579 bp encoding an inactive precursor protein with 192 amino acids. The precursor protein had no typical hydrophobic signal peptide and cleaved by interleukin-1 beta （IL-1β） converting enzyme（ICE） in caspase-1 splice site; the porcine mature protein had biological activity： After comparing with other porcine IL-18 genes, the nucleotide sequence homology was over 96% and the deduced amino acid homology was more than 98%. [ Conclusion] A full length procine IL-18 gene was gained. It lays the foundation for porcine IL-18 as an adjuvant of genetic vaccine.

摘要：Background: Diagnostic microbial isolates of bio-safety levels 3 and 4 are difficult to handle in medical field camps under military deployment settings. International transport of such isolates is challenging due to restrictions by the International Air Transport Association. An alternative option might be inactivation and sequencing of the pathogen at the deployment site with subsequent sequence-based revitalization in well-equipped laboratories in the home country for further scientific assessment. Methods: A literature review was written based on a Pub Med search. Results: First described for poliovirus in 2002, de novo synthesis of pathogens based on their sequence information has become a well-established procedure in science. Successful syntheses have been demonstrated for both viruses and prokaryotes. However, the technology is not yet available for routine diagnostic purposes. Conclusions: Due to the potential utility of diagnostic sequencing and sequence-based de novo synthesis of pathogens, it seems worthwhile to establish the technology for diagnostic purposes over the intermediate term. This is particularly true for resource-restricted deployment settings, where safe handling of harmful pathogens cannot always be guaranteed.

摘要：目的MHCⅠ类链相关基因B（MICB）是非经典的HLAⅠ类分子,可与NKG2D相互作用调节NK细胞的活性。本研究旨在建立MICB基因2～6外显子的聚合酶链反应-序列测序方法（PCR-SBT）,并分析汉族人群MICB在高分辨基因分型水平上的分布特征。方法根据GenBank数据库MICB基因有关序列（ID NC000006.11）,采用Primer3input计算机软件辅助设计引物,利用聚合酶链反应扩增MICB基因2～6外显子序列片段,扩增片段采用双酶切纯化后,按BigDye terminator v3.1sequencing Kit试剂盒操作进行测序反应,利用Assign3.5SBT分析软件指定等位基因型。
