摘要：Sedimentary pigments are useful proxy indicators of phytoplankton biomass, community structure, primary productivity and human influence in lacustrine and oceanic ecosystems. Pigments are relatively labile due to their complex chemical structures, which makes the extraction and analysis of sedimentary pigments challenging. In addition, it is important to select appropriate methods to study sedimentary pigments in regions which lack previous investigations. In this study, we adopted the L9（34） orthogonal design to develop methods of extraction and HPLC （high performance liquid chromatography） analysis of sedimentary pigments at two lakes on the Tibetan Plateau： meromictic lake-Dagze Co and dimictic lake-Jiang Co. The orthogonal design comprises 9 combinations of various parameters for extraction and HPLC analysis. The results show that the type and volume of solvent are the most important factors for pigment extraction, and the mobile phase and column selection are the most important for HPLC analysis. For the study sites, we found that the best methods to extract sedimentary pigments are as follows： the use of a mixture of acetone：methanol：water （80： 15：5, v：v：v） as the extraction solvent; sol- vent/sample ratio of 10 mL/g; sonication for 30 s and standing extraction for 6 h. The best methods for HPLC analysis are as follows： Zorbax Eclipse plus C8 column with mobile phase A, methanol：acetonitrile：0.25 M aqueous pyridine （50： 25：25, v ： v ： v） and mobile phase B, methanol： acetonitrile： acetone （20： 60： 20, v： v： v）; pH of mobile phase A adjusted to 6 with acetic acid; and HPLC column temperature maintained at 40~C. The study provides an experimental basis for future investigations of past changes in primary productivity and the response of lake ecosystems to climate change and human activities on the Tibetan Plateau.

摘要：AIM: To evaluate the biological functions of tissue-engineered human corneal epithelium (TE-HCEP) by corneal transplantation in limbal stem cell deficiency (LSCD) rabbit models. METHODS: TE-HCEPs were reconstructed with DiI-labeled untransfected HCEP cells and denuded amniotic membrane (dAM) in air-liquid interface culture, and their morphology and structure were characterized by hematoxylin-eosin (HE) staining of paraffin-sections, immunohistochemistry and electron microscopy. LSCD models were established by mechanical and alcohol treatment of the left eyes of New Zealand white rabbits, and their eyes were transplanted with TE-HCEPs with dAM surface outside by lamellar keratoplasty (LKP). Corneal transparency, neovascularization, thickness, and epithelial integrality of both traumatic and post transplantation eyes were checked once a week by slit-lamp corneal microscopy, a corneal pachymeter, and periodic acid-Schiff (PAS) staining. At day 120 post surgery, the rabbits in each group were sacrificed and their corneas were examined by DiI label observation, HE staining, immunohistochemistry and electron microscopy. RESULTS: After cultured for 5 days on dAM, HCEP cells, maintaining keratin 3 expression, reconstructed a 6-7 layer TE-HCEP with normal morphology and structure. The traumatic rabbit corneas, entirely opaque, conjunctivalized and with invaded blood vessels, were used as LSCD models for TE-HCEP transplantation. After transplantation, obvious edema was not found in TE-HCEP-transplanted corneas which became more and more transparent, the invaded blood vessels reduced gradually throughout the monitoring period. The corneas decreased to normal thickness on day 25, while those of dAM eyes were over 575 mu m in thickness during the monitoring period. A 45 layer of epithelium consisting of TE-HCEP originated cells attached tightly to the anterior surface of stroma was reconstructed 120 days after TE-HCEP transplantation, which was similar to the normal control eye

摘要：Marine in situ testing is a necessary step for stereotyping newly developed marine sensors. The use of test sites in the Yangtze Estuary area, which has high turbidity and abundant nutrients, can effectively reduce the needed testing time owing to its harsh conditions. Five test stations were established, and a floating buoy and fixed test equipment were designed. A control system, including a sensor connection, data processor, video remote transmission, and corresponding control algorithm, was developed. The control system enabled the nondestructive monitoring of biological attachments and bidirectional, real-time communication between an upper server on land and the control system at the test sites. The dissolved oxygen(DO), temperature, and pH data of DOS600 and DPS600 sensors were compared with those of AP2000 sensors. Temperature recording using the DOS600 sensor was performed nearly as well as that of the AP2000 sensor. The mean DO values(standard deviations) were 8.414 mg L-1(2.068) and 6.896 mg L-1(1.235) for the DOS600 and AP2000 sensors, respectively, indicating that the DOS600 performance was unsatisfactory. The pH recording of the DPS600 was slightly worse than that of the AP2000 sensor. Experimental results showed that the DO value was more easily affected by the buoy movement of waves compared to the pH and temperature. Moreover, data fluctuations showed that the DO and pH parameters were more vulnerable to biofouling than temperature. Waves and biofouling create a harsh test environment, and the performance difference between the developed sensors and a standard sensor can be obtained in a short time period.

摘要：In order to propose a seismic design spectrum that includes the effect of rupture directivity in the near-fault region, this study investigates the application of equivalent pulses to the parameter attenuation relationships developed for near-fault, forward-directivity motions. Near-fault ground motions are represented by equivalent pulses with different waveforms defined by a small number of parameters （peak acceleration, A, and velocity V; and pulse period, Tv）. Dimensionless ratios between these parameters （e.g., ATv/V, VTv/D） and response spectral shapes and amplitudes are examined for different pulses to gain insight on their dependence on basic pulse waveforms. Ratios of ATv/V, VTv/D, and the ratio of pulse period to the period for peak spectral velocity （Tv-p） are utilized to quantify the difference between rock and soil sites for near-fault forward-directivity ground motions. The ATv/Vratio of recorded near-fault motions is substantially larger for rock sites than that for soil sites, while Tvp/Tv ratios are smaller at rock sites than at soil sites. Furthermore, using simple pulses and available predictive relationships for the pulse parameters, a preliminary model for the design acceleration response spectra for the near-fault region that includes the dependence on magnitude, rupture distance, and local site conditions are developed.

摘要：An optimal drainage tunnel location determination method for landslide prevention was proposed to solve the existing problems in drainage tunnel construction. Current applications of drainage tunnel systems in China were reviewed and the fimctions of drainage tunnel were categorized as catchment and interception. Numerical simulations were conducted. The results show that both catchment and interception tunnels have variation of the function in the simulation of monolayer model, which shows the reduction of permeability condition in lower layer. The function of catchment can be observed in the deep slope, while the function of interception is observed near groundwater source. By using the slope safety factor and discharge water amount as the objectives of optimal drainage tunnel location, and pore-water pressure in fixed node and section flux as the judgment for construction quality of adjacent drainage tunnel, the design principle of drainage tunnel was introduced. The K103 Landslide was illustrated as an example to determine the optimal drainage tunnel location. The measured drainage tunnel efficiency was evaluated and compared with that from the numerical analyses based on groundwater data. The results validate the present numerical study.

摘要：This paper extends a prediction model for multi-directional random wave transformation based on an energy balance equation by Mase with the consideration of wave shoaling, refraction, diffraction, reflection and breaking. This numerical model is improved by 1) introducing Wen's frequency spectrum and Mitsuyasu's directional function, which are more suitable to the coastal area of China; 2) considering energy dissipation caused by bottom friction, which ensures more accurate results for large-scale and shallow water areas; 3) taking into account a non-linear dispersion relation. Predictions using the extended wave model are carried out to study the feasibility of constructing the Ai Hua yacht port in Qingdao, China, with a comparison between two port layouts in design. Wave fields inside the port for different incident wave directions, water levels and return periods are simulated, and then two kinds of parameters are calculated to evaluate the wave conditions for the two layouts. Analyses show that Layout I is better than Layout II. Calculation results also show that the harbor will be calm for different wave directions under the design water level. On the contrary, the wave conditions do not wholly meet the requirements of a yacht port for ship berthing under the extreme water level. For safety consideration, the elevation of the breakwater might need to be properly increased to prevent wave overtopping under such water level. The extended numerical simulation model may provide an effective approach to computing wave heights in a harbor.
