摘要：In this study, cinnamic acid derivatives with pyridine ring were synthesized via Hantzsch reaction in order to expand conjugation system and their antioxidant abilities were tested. According to the evaluation results of their antioxidant properties, compounds 2 and 3 have similar antioxidant activity to prevent DNA from ＊OH- and Cu2＋/GSH induced oxidation, demonstrating that 1,4-dihydropyridine and pyridine nucleus exhibit good resistance to oxidation while the substituents have little effect. In the meantime, the AAPH-induced oxidative DNA damage [AAPH=2,2＇-azobis（2-methylpropionamidine）dihydrochloride] and trapping ABTS＋＊ test show that N--H bond and large conjugation systems are the pivotal parts to improve the activities of antioxidant.

摘要：Candida albicans （C. albicans） infection has a high mortality rate in immunocompromised patients. Owing to the inefficiency of the current diagnostic system and the absence of licensed vaccines against candidiasis, the prevention of C. albicans infection remains a challenge. C. albicans infection can be evaluated and prevented by the anti-secreted aspartyl proteinase 2 antibody （anti-Sap2 IgG） and Hsp90 antibody （anti-Hsp90 IgG）. In this study, to explore a new agent for the improvement of the diagnosis and the prevention of C. albicans infection, an engineered fd bacteriophage, which is considered a human-safe virus nanofiber, was designed and prepared with two epitopes that could induce and capture anti-Sap2 IgG and anti-Hsp90 IgG. The dual-display phage was employed as a novel capture probe to develop a new enzyme-linked immunosorbent assay （ELISA） method, which significantly improved the detection rate compared with those of the ELISA in which recombinant protein Sap2 was used as coating antigen to capture the spedfic antibodies （rSap2-ELISA） and the ELISA in which recombinant protein Hsp90 was used as coating antigen to capture the specific antibodies （rHsp90-ELISA）. In addition, the nanofibers acted as a potential vaccine to immunize mice, as well as recombinant proteins, more efficiently mediated humoral and cellular immune responses, decreased levels of C. albicans colonization, and increased the survival rates in C. albicans-infected mice. Therefore, the phage dual-display nanofiber has been shown to be a powerful bifunctional agent for protection against and sensitive detection of clinical infections, which has the potential to be widely used in the life sciences, clinical medicine, and environmental sciences.

摘要：The purpose of this study was to establish a method for measuring the knee valgus angle from the ana- tomical and mechanical axes on three-dimensional reconstruction imaging models, and to use this method for estimating an average knee valgus angle value for northern Chinese adults. Computed tomographic angiography data in DICOM format for 128 normal femurs from 64 adult subjects were chosen for analysis. After the femur images were subjected to three-dimensional reconstruction, the deepest point in the intercondylar notch （point A）, the midpoint of the medullary cavity 20 cm above the knee-joint line （point B）, and the landmark of the femoral head rotation center （point C） were identified on each three-dimensional model. The knee valgus angle was defined as the angle enclosed by the distal femoral anatomical axis （line AB） and the femoral mechanical axis （line AC）. The average （mean＋SD） of knee valgus angle for the 128 femurs was 6.20°±1.20° （range, 3.05° to 10.64°）. Significant positive correlations were found between the knee valgus angles of the right and left sides and between the knee valgus angle and age. During total knee arthroplasty, choosing a valgus cut angle of approximately 6° may achieve a good result in reestablishing the natural mechanical alignment of the lower extremity for patients of northern Chinese ethnicity. Larger valgus cut angles should be chosen for older patients.

摘要：Objective： A new therapeutic strategy using nanocomposite scaffolds of grafted hydroxyapaUte （g-HA）/ poly（lactide-co-glycolide） （PLGA） carried with autologous mesenchymal stem cells （MSCs） and bone morphogenetic protein-2 （BMP-2） was assessed for the therapy of critical bone defects. At the same time, tissue response and in vivo mineralization of tissue-engineered implants were investigated. Methods： A composite scaffold of PLGA and g-HA was fabricated by the solvent casting and particulate-leaching method. The tissue-engineered implants were prepared by seeding the scaffolds with autologous bone marrow MSCs in vitro. Then, mineralization and osteogenesis were ob- served by intramuscular implantation, as well as the repair of the critical radius defects in rabbits. Results： After eight weeks post-surgery, scanning electron microscopy （SEM） and energy dispersive X-ray spectroscopy （EDX） revealed that g-HNPLGA had a better interface of tissue response and higher mineralization than PLGA. Apatite particles were formed and varied both in macropores and micropores of g-HNPLGA. Computer radiographs and histological analysis revealed that there were more and more quickly formed new bone formations and better fusion in the bone defect areas of g-HNPLGA at 2-8 weeks post-surgery. Typical bone synostosis between the implant and bone tissue was found in g-HNPLGA, while only fibrous tissues formed in PLGA. Conclusions： The incorporation of g-HA mainly im- proved mineralization and bone formation compared with PLGA. The application of MSCs can enhance bone for- mation and mineralization in PLGA scaffolds compared with cell-free scaffolds. Furthermore, it can accelerate the absorption of scaffolds compared with composite scaffolds.
